Trying to figure out rental car trip from Tokyo to Kukuna on Lake Kawaguchi.

Pretty much I am trying to figure out a rental car trip from Tokyo to Lake Kawaguchi. I was thinking of doing the public transit but with the amount of bags and time it will take, driving to the Kukuna Hotel seems the easiest.

My question is, is if I rent a car from Tokyo(most likely Toyota rentals) to Kawaguchi and return back to Tokyo; but in a different location in Tokyo as we will be getting a hotel closer to Haneda airport, will that be possible?

I know in the States we can return rental cars to different locations for an extra fee, just wondering if that is possible in Tokyo as well?
