Hey guys! Sorry we are still getting used to Japan’s transportation system. Are we able to use pasmo cards for Yokohama Eki Nishiguchi -> Osaka Eki Mae? It’s an overnight bus route on Google maps
Don’t you have to book the nightbus and pay in advance? You could use a PASMO to pay for a bus ticket at the convenience store.
PASMO and other IC cards are generally reliable for local buses, but for an overnight bus you need to book in advance. Instead of card readers long distance buses will have a passenger list. Depending on the bus you can often book until a few hours before departure, though you might miss out if it’s booked. A google search should find you some sites to book with.

As others have mentioned IC cards are not normally used for this type of bus.
However you pay, make sure to take a good look at the driver when boarding. If he looks sleepy and/or appears to be in his 70s, get the next one.
