Do you mean the “Permission to Engage in Activity other than that Permitted under the Status of Residence Previously Granted”, that lets you have a part-time job up to 28 hours per week?
If that’s it, you’d better do the paperwork before, so you can start whenever you have the chance to.
Yes, you can apply for working permission without a job lined up. IIRC you can even apply for it at the airport when you get your zairyu card.
