Things that should be done on weekdays/weekends?

Planning on visiting Tokyo, Osaka, and Kyoto. I imagine that most things should be done on weekdays to beat any rush, but are there absolutely must-do’s on weekdays? What popular touristy things can be done comfortably on weekends?
